Which one of your Black Hole Quenchers do you recommend using to label proteins?
We recommend using BHQ-10 succinimidyl ester (catalog# BHQ-10S) for the labeling of proteins. It has been shown to function as a FRET quencher for fluorophores within 100 angstroms.
How stable are the CAL Fluor and BHQ dyes when conjugated to peptides in solution?
These dyes are very stable. Their pH stability shouldn't differ much from that of the peptide in which they have been incorporated. However, BHQ's are susceptible to reduction in buffer solutions. When DTT is added to enzyme assay systems the rates of reduction vary greatly with peptide structure. Typically exposure to 50 mM DTT for 1 hour at room temperature gives <1% reduction.
